Abstract
In this work, a novel fully automated Two-Electrode Voltage-Clamp amplifier with an additional third electrode for the generation of an extracellular compensation field, has been developed in order to carry out electrophysiological experiments with oocytes of the african claw frog Xenopus laevis. Essential focus has been laid on the development of the glass micro electrodes for the injection of current and to measure membrane voltage, the design and optimization of the measurement electronics as well as the development of measurement protocols to compensate for parasitic effects like offsets and gain errors.
